Overview of PacBio HiFi Long-Read Sequencing Workflow
PacBio High-Fidelity (HiFi) Sequencing is a single-molecule, long-read sequencing technology that produces reads that are both long and accurate, using DNA fragments ranging in size from 1,000 to 20,000 bases or more. HiFi reads cover all regions of the genome, providing more complete and precise assemblies and variant calls than short-read sequencing. Unlike short reads, which can miss complex regions, HiFi sequencing captures full-length transcripts and difficult areas. Every run also includes native DNA methylation status detection, allowing the capture of sequence and epigenetic information in a single experiment without the need for additional library preparation.
PacBio Vega
The PacBio Vega is the first HiFi benchtop system and capable of delivering up to 60 Gb of long reads per run using a single SMRT Cell. This system is well-suited for custom applications of long-read sequencing, while providing the capability for whole genome, amplicon and long-read RNA sequencing.
The soon-to-be-available SPRQ-Nx chemistry will facilitate the lowering of DNA input requirements, the increase of the HiFi yield, a reduction in the run times, and drop in reagent costs.
